Specificity and Use

FOS / c-FOS antibody was raised against the antiserum was produced against a chemically synthesized phosphopeptide derived from the region of human c-Fos that contains threonine 325. The sequence is conserved in human, mouse and rat.
Human FOS / c-FOS
Optimal concentration should be evaluated by serial dilutions. For Western blotting applications, we recommend using the antibody at a 1:1000 dilution.

Nuclear phosphoprotein which forms a tight but non-covalently linked complex with the JUN/AP-1 transcription factor. In the heterodimer, FOS and JUN/AP-1 basic regions each seems to interact with symmetrical DNA half sites. On TGF-beta activation, forms a multimeric SMAD3/SMAD4/JUN/FOS complex at the AP1/SMAD-binding site to regulate TGF-beta-mediated signaling. Has a critical function in regulating the development of cells destined to form and maintain the skeleton.

Peptide Competition Lysates prepared from A431 cells left untreated (1) or treated with EGF (lanes 2 & 3) were resolved by SDS-PAGE on a 10% polyacrylamide gel and transferred to PVDF. Membranes were then left untreated (lanes 1 & 2) or treated with Lambda phosphatase (lane 3), blocked with a 3% Milk-TBST buffer for one hour at room temperature, and incubated with c-Fos [pT325] antibody for two hours at room temperature in a 3% Milk-TBST buffer. After washing, membranes were incubated with goat F(ab')2 anti-rabbit IgG HRP conjugate and bands were detected using the Pierce SuperSignal method. The data show that the phosphorylation of c-Fos on threonine 325 is induced by EGF treatment and that phosphatase treatment eliminates the signal, thereby demonstrating the phospho-specificity of the antibody.
