Specificity and Use

FAP-1 / PTPN13 antibody was raised against a synthetic peptide corresponding to amino acids 1760-1775 (EPTRQENWTPLKNDLE) of human FAP-1, GenBank no. NP_542414.1. This peptide sequence is conserved in multiple isoforms of FAP-1. Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey (100%); Marmoset, Bovine, Dog, Horse (81%).

FAP-1 typically migrates at ~270 kD on western blots. Thus for SDS-PAGE and western blot, the researcher should use techniques optimized for the transfer of proteins with high molecular weights. The actual molecular weight of FAP-1 observed depends on the isoform expression.

FAP-1 / PTPN13 is a member of the protein tyrosine phosphatase (PTP) family. PTPs are signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP is a large intracellular protein. It has a catalytic PTP domain at its C-terminus and two major structural domains: a region with five PDZ domains and a FERM domain that binds to plasma membrane and cytoskeletal elements.
