Description
ERBB3 (HER3) is a member of the epidermal growth factor receptor (EGFR) family of receptor tyrosine kinases. While ERBB3 itself is lacking kinase activity, heterodimerization of the protein with other EGF receptors leads to the activation of cell proliferation and differentiation pathways such as PI3K/Akt. It is involved in the development of the nervous system and neural crest differentiation. ERBB3 has use as a biomarker in cancer, as amplification or overexpression has been reported in prostate, bladder, breast, devil facial tumor 1 (DFT1) and other cancers and is associated with worse prognosis in solid tumors. In immunohistochemistry of normal tissue, ERBB3 has high cytoplasmic and membranous positivity in the majority of tissues. Usage
Immunohistochemistry: LS-B2126 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2126 was determined to be 5 ug/ml.
