Specificity and Use

DRD2 / Dopamine Receptor D2 antibody was raised against a 28 amino acid peptide sequence from the human D2 receptor within the cytoplasmic loop #3. The immunogen peptide shows no significant homology with other dopamine receptors (D1 and D3-D5). Species sequence Homology: Bovine, canine - 100%; Xenopus - 50%.
3rd Cytoplasmic Domain
Specific for Dopamine Receptor D2 short and long form. Species cross-reactivity: Monkey, mouse and rat.
Suitable for use in Western Blot, Immunohistochemistry, Immunoprecipitation and ELISA. Western Blot: 1:1000. Detected Dopamine Receptor D2 on 10 ug of human brain lysates. Immunohistochemistry (Paraformaldehyde fixed sections): 2-10 ug/ml. Immunoprecipitation: 1-10 ug/ml. Using 100-400 ug of protein. An antibody made to this epitope has been shown to immunoprecipitate both the long and short forms. ELISA: 0.5-1.0 ug/ml. Positive control: Monkey brain tissue, rat brain lysate.

DRD2 / Dopamine Receptor D2 is the D2 subtype of the dopamine receptor. This G-protein coupled receptor inhibits adenylyl cyclase activity. A missense mutation in this gene causes myoclonus dystonia; other mutations have been associated with schizophrenia. Alternative splicing of this gene results in two transcript variants encoding different isoforms. A third variant has been described, but it has not been determined whether this form is normal or due to aberrant splicing.
