Specificity and Use

DLL1 antibody was raised against nS0-derived rrDLL1 extracellular domain.
Rat DLL1.
Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect rat DLL1. The detection limit for rrDLL1 is approximately 5 ng/lane and 25 ng/lane under non-reducing and reducing conditions, respectively. Immunohistochemistry-This antibody has been used at a concentration of 10 µg/ml to detect DLL1 in cryosections of E13.5 mouse embryos. Tissue was fixed overnight in 4%PFA+14%picric acid in PBS at 4° C. Sections were blocked with 10% donkey serum and 0.1% Triton in 1% BSA in PBS for 45 minutes at room temperature. After blocking, sections were incubated with primary antibody for 3 hours at room temperature followed by fluorochrome-coupled anti-sheep lgG at room temperature for 1 hour. Between each step, sections were washed with 0.1 % BSA in PBS. Flow cytometry-This antibody was tested in flow cytometry using mouse splenocytes. Dilute this antibody to 25 µg/ml and add 10 pL of the diluted solution to 1-2.5 x 105 cells in a total reaction volume not exceeding 200 pL. The binding of unlabeled antibodies may be visualized by adding a secondary developing reagent such as anti-sheep antibody conjugated to a fluorochrome. Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect rat DLL1. The detection limit for rrDLL1 is approximately 0.3 ng/well. In this format, this antibody shows approximately 10% cross-reactivity with rh DLL- 1 and less than 1% cross reactivity with rrNotch-1.
