Specificity and Use

Isolated from human breast cancer carcinoma cell line MCF-7.
Recognizes all simple epithelia including glandular epithelia. Does not react with stratified squamous epithelium. Useful for the differentiation between carcinomas and lymphomas. Directed against the 52.2kD cytokeratin 8 (originally reported also as 45 kD cytokeratin 18 in the Moll-catalogue) in immunoblotting. Epitope has been mapped to aa 353-367 on alpha helical rod domain of cytokeratin 8. Does not react with mouse or rat.
Suitable for use in Immunohistochemistry, Western blot and cell suspensions. Immunohistochemistry: 1:5-1:10 frozen and paraffin-embedded tissue require Proteolytic treatment with pepsin or pronase.

About Cytokeratin 8+18

Keratin 18 is a type I cytokeratin. It is, together with its filament partner keratin 8, perhaps the most commonly found products of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene.
