Specificity and Use

CSF2 / GM-CSF antibody was raised against e. coli-derived recombinant human GMCSF Ala18-Glu144 Accession # P04141.
Detects human GMCSF in Western blots.
Western Blot, 1 µg/ml, Recombinant Human GM-CSF. under non-reducing conditions only. Immunocytochemistry, 8-25 µg/ml. Neutralization, Measured by its ability to neutralize GM-CSF-induced proliferation in the TF-1 human erythroleukemic cell line. The Neutralization Dose (ND50) is typically 0.3-0.5 µg/ml in the presence of 0.5 ng/ml Recombinant Human GM-CSF.

CSF2 / GM-CSF is a cytokine that controls the production, differentiation, and function of granulocytes and macrophages. The active form of the protein is found extracellularly as a homodimer. This gene has been localized to a cluster of related genes at chromosome region 5q31, which is known to be associated with interstitial deletions in the 5q- syndrome and acute myelogenous leukemia. Other genes in the cluster include those encoding interleukins 4, 5, and 13.


GMCSF in Human PBMCs. Granulocyte Macrophage Colony Stimulating Factor (GM-CSF) was detected in human peripheral blood mononuclear cells (PBMCs) using Human GM-CSF Monoclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using NorthernLights 557-conjugated Anti-Mouse IgG Secondary Antibody (red) and counterstained with DAPI (blue).


Cell Proliferation Induced by GM-CSF and Neutralization by Human GM-CSF Antibody. Recombinant Human GM-CSF stimulates proliferation in the TF-1 human erythroleukemic cell line in a dose-dependent manner (orange line). Proliferation elicited by Recombinant Human GM-CSF (0.5 ng/ml) is neutralized (green line) by increasing concentrations of Human GM-CSF Monoclonal Antibody. The ND50 is typically 0.3-0.5 ug/ml.
