Specificity and Use

Complement C3d antibody was raised against human C3d
Human Complement C3d. Tested in immunoelectrophoresis and double radial immunodiffusion against normal human serum, a single precipitin line is obtained and giving a reaction of full identity with the purified immunogen. The antiserum is also reacting with fragments bearing common determinants: C3, C3b, C3bi. The product does not react with any other proteins component of human serum or plasma. In supernatants of sera containing 10% polyethyleneglycol only a reaction with C3d has been obtained. Cross-reactivity Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins, since homologous proteins of different species frequently share antigenic determinants. Cross-reactivity of this antiserum has not been tested in detail.
This product is intended for use in precipitating techniques as immunoelectrophoresis and single and double radial immunodiffusion (Mancini, Ouchterlony) to identify the presence of complement C3d or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included. Determinations of individual complement components can be very useful in defining the exact location of a defect.

About Complement C3d

Complement C3 plays a central role in the activation of complement system. Its activation is required for both classical and alternative complement activation pathways. One form of C3-convertase, also known as C4b2a, is formed by a heterodimer of activated forms of C4 and C2. It catalyzes the proteolytic cleavage of C3 into C3a and C3b, generated during activation through the classical pathway as well as the lectin pathway.
