Specificity and Use

Collagen Type II from human knee cartilage and bovine nasal cartilage.
Anti-Collagen antibodies have been used for indirect trapping ELISA for quantitation of antigen in serum using a standard curve, for Immunoprecipitation and for Western Blot for highly sensitive qualitative analysis. This product was assayed by immunoblot and found to be reactive against Collagen II at a dilution of 1:5000 to 1:10000. This product was also assayed against 1.0 ug of Collagen II in a standard sandwich ELISA using Peroxidase conjugated Affinity Purified anti-Rabbit IgG [H&L] (Goat) (catalog no. LS-C60884) and ABTS as a substrate for 30 minutes at room temperature. A working dilution of 1:4000 to 1:8000 of the stock concentration is suggested for this product. For immunohistochemistry on frozen tissue sections dilute the product 1:50 to 1:200.


0.125 M sodium borate, 0.075 M sodium chloride, 0.005 M EDTA;, pH 8.0, 0.01% sodium azide.
Store vial at 4 C prior to opening. This product is stable at 4 C as an undiluted liquid. Dilute only prior to immediate use. For extended storage mix with an equal volume of glycerol, aliquot contents and freeze at -20 C or below.

About Collagen II

Type II collagen is the basis for articular cartilage and hyaline cartilage. It makes up 50% of all protein in cartilage and 85-90% of collagen of articular cartilage. Type II collagen does form fibrils. This fibrillar network of collagen allows cartilage to entrap the proteoglycan aggregate as well as provide tensile strength to the tissue.


Anti-Collagen II Antibody - Immunohistochemistry. anti-collagen II antibody (1:400, 45 min RT) showed moderate staining of in FFPE human bronchiolar cartilage (shown in image). Though not shown, faint to moderate staining of tonsillar squamous epithelium, prostatic stroma, breast, colon, placenta, and dermal connective tissues was also observed. All other tissues, including brain, breast epithelium, colon epithelium, heart, intestine, kidney, liver, lung, skeletal muscle, pancreas, spleen, testis, thymus, thyroid, and uterus were negative for staining Slides were steamed in 0.01 M sodium citrate buffer, pH 6.0 at 99-100°C - 20 minutes for antigen retrieval.
