Immunohistochemical analysis of CMKLR1 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

Specificity and Use

Peptide corresponding to an epitope within the N-terminal region of human CMKLR1.
N-Terminus
Recognizes human serpentine chemokine-like receptor 1 (CMKLR1), a novel chemoattractant receptor, which is also referred to Chem23 or DEZ. CMKLR1 is expressed on plasmacytoid dendritic cells (pDC) in blood but is not expressed on myeloid dendritic cells, lymphocytes, monocytes or granulocytes. Studies show that CMKLR1 is also expressed on dendritic cells, which have been generated in vitro from monocytes. The sole ligand for CMKLR1 is chemerin, a chemoattractant that exists at the mRNA level in a wide range of tissues, and as an inactive proform in serum and plasma. Studies suggest that chemerin, in plasma, is converted into a potent pDC attractant following blood coagulation and may be a process by which pDC's are recruited to sites of bleeding and tissue damage.

Receptor for the chemoattractant adipokine chemerin/RARRES2 and for the omega-3 fatty acid derived molecule resolvin E1. Interaction with RARRES2 induces activation of intracellular signaling molecules, such as SKY, MAPK1/3 (ERK1/2), MAPK14/P38MAPK and PI3K leading to multifunctional effects, like, reduction of immune responses, enhancing of adipogenesis and angionesis. Resolvin E1 down-regulates cytokine production in macrophages by reducing the activation of MAPK1/3 (ERK1/2) and NF-kappa-B.
