Specificity
Recognizes p27 when it is phosphorylated at the Ser10 residue. Lysates of HEK 293T cells transfected with an expression plasmid encoding a p27 mutant where Ser10 was substituted with an alanine residue were used to confirm the antibody's specificity. Reactivity was confirmed with HEK293 cells expressing wild-type p27/KIP that is normally phosphorylated. NIH3T3 and MCF-7 cell lysates can also be used as positive controls. Species cross-reactivity: Human, rat and mouse.
Usage
Suitable for use in Western Blot, Immunoprecipitation and Immunofluorescence. Western Blot: 1-3 ug/ml. Immunoprecipitation: 5-10 ug/IP reaction. Immunofluorescence: 5-10 ug/ml.

About CDKN1B / p27 Kip1
Important regulator of cell cycle progression. Involved in G1 arrest. Potent inhibitor of cyclin E- and cyclin A-CDK2 complexes. Forms a complex with cyclin type D-CDK4 complexes and is involved in the assembly, stability, and modulation of CCND1-CDK4 complex activation. Acts either as an inhibitor or an activator of cyclin type D-CDK4 complexes depending on its phosphorylation state and/or stoichometry. P46527 NM_004064 NP_004055.1
