Mouse recombinant p27Kip1 protein.
Highly specific for p27kip1 (27kD). Does not cross-react with other closely related mitotic inhibitors. Co-precipitates cdk4 in complex p27Kip1. Its epitope is mapped between aa83-204 of p27. Species cross-reactivity: Human, monkey, canine, mouse and rat. Does not cross-react with bovine.
Suitable for use in Flow Cytometry, Immunofluorescence, Western Blot, Immunoprecipitation and Immunohistochemistry. Western Blot: 1:100-1:200. Immunohistochemistry: 1:25-1:50 (Frozen, formalin/methacarn, and paraffin embedded sections). Note: Staining of formalin/paraffin sections requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 minutes, followed by cooling at room temperature for 20 minutes. Immunoprecipitation: 10 ul/mg of protein lysate. Positive control: HeLa cells, MAD 109 cells, Ls174T cells, breast carcinoma or colon carcinoma.
Important regulator of cell cycle progression. Involved in G1 arrest. Potent inhibitor of cyclin E- and cyclin A-CDK2 complexes. Forms a complex with cyclin type D-CDK4 complexes and is involved in the assembly, stability, and modulation of CCND1-CDK4 complex activation. Acts either as an inhibitor or an activator of cyclin type D-CDK4 complexes depending on its phosphorylation state and/or stoichometry.
