Specificity and Use

CDH6 / K Cadherin antibody was raised against nS0-derived rhCadherin-6 extracellular domain.
Human Cadherin-6 extracellular domain.
In direct ELISAs and Western blots, this antibody shows less than 5% cross-reactivity with rhCadherin-8,. rhCadherin-11, and rhCadherin-12. Applications. Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect human Cadherin-6. The detection limit for rhCadherin-6 is approximately 10 ng/lane under non-reducing and reducing conditions. Immunocytochemistry-This antibody has been used at a concentration of 10 µg/ml to detect human Cadherin-6 in MG63 human osteosarcoma cells cells. Cells were fixed with PBS containing 4% paraformaldehyde for. 20 minutes at room temperature and blocked with PBS containing 10% normal donkey serum, 0.1 % Triton X-100, and 1 % BSA for 45 minutes at room temperature. After blocking, cells were incubated with diluted primary antibody for 3 hours at room temperature followed by donkey anti-sheep lgG at room temperature for 1 hour. Between each step, cells were washed with PBS containing 0.1% BSA. Flow cytometry-This antibody has been tested on MG63 human osteosarcoma cells for use in flow cytometry. Dilute this antibody to 50 µg/ml and add 10 ul of the diluted solution to 1-2.5 x 105 cells in a total reaction volume not exceeding 200 ul. The binding of unlabeled antibodies may be visualized by adding 10 ul of a 25 µg/ml stock solution of a secondary developing reagent such as anti-sheep lgG conjugated to a fluorochrome. Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect human Cadherin-6. The detection limit for rhCadherin-6 is approximately 1 ng/well.
