Specificity and Use

CD22 antibody was raised against nS0-derived rmSiglec-2 extracellular domain.
Mouse Siglec-2 extracellular domain.
Neutralization of mouse Siglec-2 bioactivity. The exact concentration of antibody required to neutralize rmSiglec-2 activity is dependent on the cytokine concentration, cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the cytokine activity on a responsive cell line, when that cytokine is present at a concentration just high enough to elicit a maximum response. Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect mouse Siglec-2. The detection limit for rmSiglec-2 is approximately. 5 ng/lane under non-reducing and reducing conditions. Immunocytochemistry-This antibody will detect Siglec-2 in cells. The working dilution is 2-15 µg/ml. Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect mouse Siglec-2. The detection limit for rmSiglec-2 is approximately. 0.2 ng/well. In this format, this antibody shows approximately 20% cross-reactivity with rhSiglec-2 and less than 1 % cross-reactivity with rhSiglec-5, rhSiglec-7, rhSiglec-9, rhSiglec-10, rmSiglec-3 and rmSiglec-F.

Mediates B-cell B-cell interactions. May be involved in the localization of B-cells in lymphoid tissues. Binds sialylated glycoproteins; one of which is CD45. Preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site can be masked by cis interactions with sialic acids on the same cell surface. Upon ligand induced tyrosine phosphorylation in the immune response seems to be involved in regulation of B-cell antigen receptor signaling.
