The scaffolding protein encoded by this gene is the main component of the caveolae plasma membranes found in most cell types. The protein links integrin subunits to the tyrosine kinase FYN, an initiating step in coupling integrins to the Ras-ERK pathway and promoting cell cycle progression. The gene is a tumor suppressor gene candidate and a negative regulator of the Ras-p42/44 mitogen-activated kinase cascade.
Caveolin-1 in Human Liver. Caveolin-1 was detected in immersion fixed paraffin-embedded sections of human liver using Human Caveolin-1 Antigen Affinity-purified Polyclonal Antibody at 10 ug/ml overnight at 4°C. Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic. Tissue was stained using Anti-Goat HRP-DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to endothelial cells in bile canaliculi.
Detection of Human Caveolin-1 by Western Blot. Western blot shows lysates of HUVEC human umbilical vein endothelial cells, A431 human epithelial carcinoma cell line, and A549 human lung carcinoma cell line. PVDF Membrane was probed with 0.2 ug/ml of Human Caveolin-1 Antigen Affinity-purified Polyclonal Antibody followed by HRP-conjugated Anti-Goat IgG Secondary Antibody. Specific bands were detected for Caveolin-1 at approximately 21 to 24 kD (as indicated). This experiment was conducted under reducing conditions.
