Specificity and Use

CASP9 / Caspase 9 antibody was raised against the N-terminal fragment of human Caspase 9 (amino acid residues 1-134).
N-Terminus
Recognizes the proform and the active cleaved form of Caspase 9, Mr 46kD and 34kD, respectively. Does not recognize Caspase 3, 6, 7 and 8. Species cross-reactivity: Human, not mouse or rat.
Suitable for use in ELISA and Western Blot. Western Blot Analysis: 0.5-2 ug/ml detects the proform of Caspase 9 in RIPA lysates from both A431 and Raji cells. 0.5-2 ug/ml of a previous lot detected the proform and cleaved Caspase 9 in RIPA lysates from HFF cells. A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Caspase 9 (2 ug/ml). Proteins were visualized using a goat antimouse secondary antibody conjugated to HRP and a chemiluminescence detection system.

100 ul of 0.1 M Tris-glycine, pH 7.4, 0.15 M sodium chloride, 0.05% sodium azide, 30% glycerol.

CASP9 / Caspase 9 is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme.
