Specificity and Use
CASP8 / Caspase 8 antibody was raised against peptide sequence corresponding to the C-terminus of human Caspase-8 (PQPTFTLRKKLVFPSD). This sequence is common to various Caspase-8 forms including Caspase 8a (aa481-496), Caspase 8b (aa464-479), Caspase 8c (aa449-464), Caspase8L (aa523-538), Truncated caspase 8 (as251-266). Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey (100%); Marmoset (94%); Orangutan, Mouse, Bovine, Dog, Bat, Elephant, Panda, Horse, Pig (81%).

Most upstream protease of the activation cascade of caspases responsible for the TNFRSF6/FAS mediated and TNFRSF1A induced cell death. Binding to the adapter molecule FADD recruits it to either receptor. The resulting aggregate called death-inducing signaling complex (DISC) performs CASP8 proteolytic activation. The active dimeric enzyme is then liberated from the DISC and free to activate downstream apoptotic proteases.
