Specificity and Use

CASP3 / Caspase 3 antibody was raised against synthetic peptide (KLH coupled) corresponding to amino-terminal residues adjacent to (Asp175) in human caspase-3.
N-Terminus
Detects endogenous levels of the large fragment (17/19kD) of activated caspase-3 resulting from cleavage adjacent to Asp175. Does not recognize full length caspase-3 or other cleaved caspases.
Suitable for use in Flow Cytometry, Western Blot, Immunoprecipitation, Immunohistochemistry, Immunofluorescence. Western Blot: 1:1000. Immunoprecipitation: 1:50. Immunohistochemistry (Frozen): 1:200 using 3% formaldehyde as fixative. Immunohistochemistry (Paraffin): 1:200. Antigen retrieval: heat in citrate buffer, pH 8.0. Immunofluorescence (IC): 1:200. Flow Cytometry: 1:200.

10 mM sodium HEPES, pH 7.5, 150 mM sodium chloride, 0.1 mg/ml BSA, <0.02% sodium azide, 50% glycerol.

Caspase 3 is synthesized as an inactive proenzyme that undergoes proteolytic cleavage to produce 2 subunits, termed p20 and p11. These subunits dimerize to form the active enzyme. Caspase 3 proteolytically cleaves and activates other proteins. Caspase 3 has been associated with apoptosis.
