Services
  • Immunohistochemistry Services

  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

c-Kit / CD117 antibody was raised against the antiserum was produced against a chemically synthesized phosphopeptide derived from the region of human c-Kit that contains tyrosine 721. (Mouse, Rat have 82% homology and are expected to work but have not been tested).

Optimal concentration should be evaluated by serial dilutions. For Western blotting applications, we recommend using the antibody at a 1:1000 dilution.

Packaging

PBS, pH 7.3, 1.0 mg/ml BSA, 50% glycerol, 0.05% sodium azide
Store at -20°C. Aliquot to avoid freeze/thaw cycles.

KIT Antibody, C-Kit Antibody, CD117 Antibody, PBT Antibody, Piebald trait Antibody, Piebald trait protein Antibody, Proto-oncogene c-Kit Antibody, SCFR Antibody, Soluble KIT variant 1 Antibody, Tyrosine-protein kinase Kit Antibody, p145 c-kit Antibody, CD117 antigen Antibody

Tyrosine-protein kinase that acts as cell-surface receptor for the cytokine KITLG/SCF and plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. In response to KITLG/SCF binding, KIT can activate several signaling pathways. Phosphorylates PIK3R1, PLCG1, SH2B2/APS and CBL.

Peptide Competition and Stimulation Extracts prepared from Mo7e cells left untreated (1) or treated with 100 ng/mL human SCF for 5 minutes (2-5) were resolved by SDS-PAGE on a 10% polyacrylamide gel and transferred to PVDF. The membrane was blocked with a 5% BSA-TBST buffer overnight at 4oC, then incubated with the c-Kit [pY721] antibody for two hours at room temperature in a 3% BSA-TBST buffer, following prior incubation with: no peptide (1, 2), the non-phosphopeptide corresponding to the phosphopeptide immunogen (3), a generic phosphotyrosine-containing peptide (4), or the phosphopeptide immunogen (5). After washing, the membrane was incubated with goat F(ab')2 anti-rabbit IgG alkaline phosphatase and signals were detected using the Tropix WesternStar method. The data show that only the phosphopeptide corresponding to c-Kit [pY721] blocks the antibody signal, demonstrating the specificity of the antibody. The data also show the induction of c-Kit [pY721] phosphorylation by the addition of SCF to this cell system.
