Specificity and Use

c-CBL antibody was raised against gST fusion protein corresponding to residues 878-906 of human Cbl. This sequence is identical in mouse.

Specific for Cbl. Species cross-reactivity: mouse and rat.
Suitable for use in Western Blot, Immunoprecipitation, and Immunocytochemistry. Western Blot Analysis: 0.5-2 ug/ml detects Cbl (120kD) in RIPA lysates from human Jurkat and mouse 3T3 cells. 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with anti-Cbl (1 ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 2 ug of a previous lot immunoprecipitated Cbl from 1 ug of a mouse 3T3 cell RIPA lysate. Immunocytochemistry: 5-10 ug/ml of a previous lot showed positive immunostaining for Cbl in 3T3 cells fixed with ice cold methanol/ethanol [1:1] for 1 minute. Positive control: F4209:3T3 lysate. Use 20 ug per lane for minigels.

Adapter protein that functions as a negative regulator of many signaling pathways that are triggered by activation of cell surface receptors. Acts as an E3 ubiquitin-protein ligase, which accepts ubiquitin from specific E2 ubiquitin-conjugating enzymes, and then transfers it to substrates promoting their degradation by the proteasome. Recognizes activated receptor tyrosine kinases, including KIT, FLT1, FGFR1, FGFR2, PDGFRA, PDGFRB, EGFR, CSF1R, EPHA8 and KDR and terminates signaling.
