Specificity and Use

BIRC7 / Livin antibody was raised against a synthetic peptide corresponding to amino acids 199-217 (PELPTPRREVQSESAQEPG) of human Livin, isoform alpha and isoform beta, GenBank nos. NP_647478 (Livin, isoform alpha) and NP_071444.1 (Livin, isoform beta). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Gibbon, Monkey (100%); Orangutan (95%); Marmoset (84%); Rat (68%).

Livin is a short-lived protein that is thought to be degraded by the ubiquitin-proteasome pathway. Thus users may want to do a time course when examining the effects of treatments on Livin expression. Users are encouraged to refer to the literature for additional information; for example, Ma et al. 2006. 2. Livin has been shown to undergo proteolytic cleavage during apoptosis. Thus the molecular weight of Livin observed on Western blots may vary depending on the presence of absence of cleavage fragments.

Apoptotic regulator capable of exerting proapoptotic and anti-apoptotic activities and plays crucial roles in apoptosis, cell proliferation, and cell cycle control. Its anti-apoptotic activity is mediated through the inhibition of CASP3, CASP7 and CASP9, as well as by its E3 ubiquitin-protein ligase activity. As it is a weak caspase inhibitor, its anti-apoptotic activity is thought to be due to its ability to ubiquitinate DIABLO/SMAC targeting it for degradation thereby promoting cell survival.
