Specificity and Use
BCL2 / Bcl-2 antibody was raised against synthetic peptide corresponding to residues 41-54 (Cys-GAAPAPGIFSSQPG) of human bcl-2 protein. Cellular Localization: Cytoplasm. MW of antigen: 25-26kD. Percent identity by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Dog, Cat, Pig (100%); Bovine (93%); Horse (86%).
Recognizes human Bcl-2a at ~25kD. Species cross-reactivity: monkey. Does not react with mouse or rat.
Immunohistochemistry: LS-B2352 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2352 was determined to be 5 ug/ml. Western Blot: 1 ug/ml for 2hrs at RT. Positive control: Tonsil or follicular lymphomas. Raji or HeLa cells.

Suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. Regulates cell death by controlling the mitochondrial membrane permeability. Appears to function in a feedback loop system with caspases. Inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1).
