  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

BBC3 / PUMA antibody was raised against synthetic peptide ARARQEGSSPEPVEG corresponding to the N-terminus of human PUMA alpha. Percent identity by BLAST analysis: Human, Gibbon, Mouse, Rat, Porcine (100%); Opossum (87%).
Recognizes the p53 - upregulated modulator of apoptosis, also known as Bcl-2 binding component 3, JFY-1 or Bcl-2. PUMA is a 193 amino acid ~20 kD pro-apoptotic member of the BCL-2 family. Bcl-2 binding component 3 is localized to the mitochondria, binds BCL-2 and induces cytochrome C release and apoptosis in response to p53. Four isoforms of PUMA have been described due to alternative splicing. Is expected to recognize the canonical isofom 1, also known as PUMA alpha, it is not expected to recognize any of the other isoforms of PUMA.
Immunohistochemistry: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH6.0 is recommended for this purpose. Histology Positive Control Tissue: Human breast carcinoma. Western Blot: detects a band of approximately 23kD in K562 cell lysates.

PBS, 0.02% sodium azide.
Short term 4°C, long term aliquot and store at -20°C, avoid freeze thaw cycles.

About BBC3 / PUMA

BBC3 Antibody, Bcl-2-binding component 3 Antibody, BCL2 binding component 3 Antibody, JFY-1 Antibody, JFY1 Antibody, PUMA Antibody

Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes. A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) and bbc3 (for Bcl-2 binding component 3) in human and mouse. PUMA/bbc3 is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53.

Anti-BBC3 / PUMA antibody IHC of human colon. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody LS-B7285 concentration 2.5 ug/ml.
