Specificity and Use

BARD1 antibody was raised against synthetic peptide corresponding to a region of human BARD1 encoded within exon 4.
Human aurora A kinase, also known as Serine/Threonine Kinase 6, STK6, Breast tumor-amplified kinase, ARK-1, Serine/threonine-protein kinase 15 or Aurora/IPL1-related kinase 1. Aurora A kinase is a cell-cycle regulated kinase that has an important role in centrosome maturation and chromosome segregation. This protein is found at the centrosome in interphase cells and at the spindle poles in mitosis. Aurora A kinase is frequently overexpressed in various human carcinomas, which would suggest that this protein may be involved in tumorigenesis (Bischoff et al. 1998).

BARD1 is a protein which interacts with the N-terminal region of BRCA1. In addition to its ability to bind BRCA1 in vivo and in vitro, it shares homology with the 2 most conserved regions of BRCA1: the N-terminal RING motif and the C-terminal BRCT domain. The RING motif is a cysteine-rich sequence found in a variety of proteins that regulate cell growth, including the products of tumor suppressor genes and dominant protooncogenes. This protein also contains 3 tandem ankyrin repeats.

Detection of human BARD1 in nuclear extracts of HeLa and U2OS cells and in U2OS cells either expressing endogenous (-) or over-expressing (+) human BARD1 by Western blotting with Rabbit anti-Human BARD1 (LS-C43051). Immunoprecipitation of human Bard1 from nuclear extracts of HeLa cells using AHP871 (2 ug/100ug lysate) or normal rabbit IgG as a control.
