Specificity and Use
BAG3 / BAG-3 antibody was raised against a synthetic peptide corresponding to amino acids 276-294 (REGSPARSSTPLHSPSPIR) of human BAG-3, GenBank no. gi|14043024|ref|NP_004272.2| The amino acid sequence used for immunogen is 84% conserved with rat and mouse BAG-3. Percent identity by BLAST analysis: Human, Gibbon, Monkey, Marmoset (100%); Gorilla (95%); Hamster, Elephant, Horse (89%); Mouse, Rat, Panda, Rabbit (84%).

BAG proteins compete with Hip for binding to the Hsc70/Hsp70 ATPase domain and promote substrate release. All the BAG proteins have an approximately 45-amino acid BAG domain near the C terminus but differ markedly in their N-terminal regions. The protein encoded by this gene contains a WW domain in the N-terminal region and a BAG domain in the C-terminal region. The BAG domains of BAG1, BAG2, and BAG3 interact specifically with the Hsc70 ATPase domain in vitro and in mammalian cells.
Western blot of BAG-3 in CAPAN-1 human pancreatic cancer cells following heat shock using LS-C148217 at !:2000, Cells were heat shocked at 45 degrees for 10min and total proteins were extracted following incubation at 37 degrees for the indicated times. Control= no heat shock treatment. Maxi mal BAG-3 protein expression was detected at 6 h post heat shock treatment.
