Specificity and Use

Alpha-1-Antichymotrypsin antibody was raised against native, purified.
Recognizes erythroid ankyrin (ankyrin-1), also known as ankyrin R or erythrocyte ankyrin. Ankyrin 1 is a 1881 amino acid ~220kD cytoplasmic protein forming an attachment for a number of integral membrane proteins and cytoskeletal elements. Ankyrin-1 contains a large number of repeats, a domain and two domains. Multiple isoforms of ankyrin-1 are generated by alternative splicing and promoter usage,is expected to recognize all erythrocyte isoforms of ankyrin-1 but is not expected to recognize the N-terminally truncated muscle isoforms of ankyrin-1. Additionallyis likely to cross react with ankyrin-1 from chimpanzee (Pan troglodytes) and lowland gorilla (Gorilla gorilla gorilla). Most ankyrins are composed of 3 structural domains; a N-terminal regulatory domain containing multiple ankyrin repeats, a central spectrin binding domain and a C-terminal regulatory domain. Ankyrin-1 is present in erythrocytes and has also been found in brain and muscle. Mutations in erythrocytic ankyrin-1 are associated with hereditary spherocytosis.
The applications listed have been tested for the unconjugated form of this product. Other forms have not been tested.

Alpha-1-Antichymotrypsin is a plasma protease inhibitor and member of the serine protease inhibitor class. Polymorphisms in this protein appear to be tissue specific and influence protease targeting. Variations in this protein's sequence have been implicated in Alzheimer's disease, and deficiency of this protein has been associated with liver disease. Mutations have been identified in patients with Parkinson disease and chronic obstructive pulmonary disease.
