Specificity and Use

SLPI antibody was raised against purified, E. coli-derived, recombinant human secretory leukocyte protease inhibitor (SLPI).
Recognizes human Secretory Leukocyte Protease Inhibitor (SLPI).
Immunohistochemistry: LS-B2174 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2174 was determined to be 2.5 ug/ml. Neutralization: 0.7ug will neutralize 50% of the activity due to 1ng of rhSLPI. Inhibits trypsin-catalyzed hydrolysis of tosyl-Gly-Pro-Lys-4-nitroanilid acetate. Recombinant human SLPI will inhibit trypsin activity at a 1:1 molar ratio of rhSLPI to active trypsin.Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~1ng/well.Western Blot: 1-2ug/ml. The detection limit is ~0.2ng/lane under non-reducing conditions. Because this antibody is a preparation is a total IgG fraction, complete monospecificity cannot be assumed.
