Anti-AKT Monoclonal Antibody - Western Blot. anti-AKT Monoclonal Antibody in western blot shows detection of AKT (indicated by arrowhead at ~56 kD) on unstimulated (lane 1) and PDGF stimulated NIH/3T3 cell lysates (lane 2). Each lane contained approximately 10 ug of lysate. All samples were loaded on to a 4-20% gradient gel for separation. After electrophoresis, the gel was blocked with 5% BLOTTO (B501-0500 in TBS for 2h at RT. The membrane was probed with Anti-AKT antibody at a 1:1000 dilution in TBS with 5% BLOTTO, overnight at 4° C. For detection HRP conjugated Gt-a-Mouse IgG (p/n LS-C60680) was used at a 1:40000 dilution for 1 h at 4° C with FemtoMax enhanced chemiluminescent reagent (p/n FEMTOMAX-100). Images were captured using 2X2 binning for 10-20 sec using a BioSpectrum Imaging System (UVP Ltd.).

About AKT1
AKT1 is one of 3 closely related serine/threonine-protein kinases (AKT1, AKT2 and AKT3) called the AKT kinase, and which regulate many processes including metabolism, proliferation, cell survival, growth and angiogenesis. This is mediated through serine and/or threonine phosphorylation of a range of downstream substrates. Over 100 substrate candidates have been reported so far, but for most of them, no isoform specificity has been reported. P31749 NM_005163 NP_005154.2

Usage
Immunohistochemistry: LS-B1327 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1327 was determined to be 5 ug/ml.
