Specificity and Use

Synthetic peptide from human AKT (AKT1/AKT2/AKT3).
C-Terminus
Synthetic peptide C-R-P-H-F-P-Q-F-S-Y-S-A-S-G-T-A corresponding to the C-terminus (460-480) of human, rat and mouse and chicken AKT proteins conjugated to KLH using maleimide. A residue of cysteine was added to the amino terminal end to facilitate coupling.
Immunohistochemistry: LS-B2844 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2844 was determined to be 1:100.

Packaging

0.02 M potassium phosphate, 0.15 M sodium chloride, pH 7.2, 0.1% sodium azide.
Store vial at -20 C prior to opening. Dilute only prior to immediate use. For extended storage aliquot contents and freeze at -20 C or below. Avoid cycles of freezing and thawing.

About AKT1 + AKT2 + AKT3

AKT, also known as protein kinase B (PKB), constitutes a family of serine/threonine kinases. AKT has 3 isoforms: AKT1, AKT2), and AKT3). These isoforms share a high degree of homology, especially in the catalytic domains (87%-90% identical amino acids).

Secondary antibody: HRP conjugated Gt-a-Rabbit IgG (LS-C60865) at 1:10000 preceded color development using Pierce Chemical's SuperSignal substrate. Block: MOPS buffer overnight at 4C. Predicted/Observed size: 128 kDa, 128 kDa for AKT. Other band(s): none.
