Services
  • Immunohistochemistry Services

  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

ADRA2A antibody was raised against (C)KASRWRGRGNREKR; Cysteine added for conjugation. Percent identity by BLAST analysis: Human, Gorilla, Monkey, Mouse, Rat, Elephant, Panda, Cat, Bovine, Pig, Opossum, Guinea pig, Lizard (93%); Pufferfish, Zebrafish (86%).
Recognizes rat alpha 2a Adrenergic Receptor. Shows positive staining in rat spinal cord dorsal horn, ventral horn motor neurons and retina. When examined in other brain regions known to express alpha 2a, staining was negative. Species cross-reactivity: Mouse and human.
Suitable for use in Immunohistochemistry and Immunocytochemistry. Immunohistochemistry: 1:100-1:500; Antiserum was used on perfusion fixed tissue. Perfusion: 1) Calcium-free Tyrode's solution, 2) paraformaldehyde-picric acid fixative, and 3) 10% sucrose in PBS as a cryo-protectant. Desired tissues were dissected and stored overnight in 10% sucrose in PBS. Slide mounted tissue sections were processed for indirect immunofluorescence. Slides were incubated with blocking buffer for 1 hour at room temperature. Primary antiserum was diluted with blocking buffer to the appropriate working concentration. Blocking buffer was removed and slides were incubated for 18-24 hours at 4?C with primary antiserum. Slides were rinsed 3 times and then incubated with secondary antibodies for 1 hour at room temperature. Slides were again rinsed 3 times and cover-slipped. Staining was examined using fluorescence microscopy.

Alpha-2-adrenergic receptors are members of the G protein-coupled receptor superfamily. They include 3 highly homologous subtypes: alpha2A, alpha2B, and alpha2C. These receptors have a critical role in regulating neurotransmitter release from sympathetic nerves and from adrenergic neurons in the central nervous system.
