Specificity and Use

RELA / NFKB p65 antibody was raised against synthetic peptide from human RELA.

Immunohistochemistry: LS-B1488 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1488 was determined to be 10 ug/ml.

NFKB1 or NFKB2 bind to REL, RELA, or RELB to form the NFKB complex. The NFKB1/RELA heterodimer is the most abundant form of NFKB. The NFKB complex is inactivated by I-kappa-B proteins that keep it trapped in the cytoplasm. Phosphorylation of serine residues on the I-kappa B proteins mark them for ubiquitination and destruction, and NFKB translocates to the nucleus where it binds DNA.
