Description
NOTCH1 is a transmembrane receptor that binds Jagged1, Jagged2 and Delta1 to regulate cell-fate determination. NOTCH1 affects the implementation of differentiation, proliferation and apoptotic programs. These include a number of functions in the brain, such as promoting progenitor cell differentiation into astrocytes and repressing neuronal and myogenic differentiation. NOTCH1 is also involved in angiogenesis, and negatively regulates endothelial cell proliferation and migration and angiogenic sprouting. During cerebellar development, NOTCH1 functions as a receptor for neuronal DNER and is involved in the differentiation of Bergmann glia. In immunohistochemistry, NOTCH1 has positivity in most tissues throughout the body. Anti-Notch antibody was prepared by repeated immunizations with a synthetic peptide corresponding to an internal region near amino acid 2480-2510 of human Notch 1.  A residue of cysteine was added to the amino terminal end to facilitate coupling.

Usage
Immunohistochemistry: LS-B398 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B398 was determined to be 1:500.
