SHP2, a NT2 non-receptor protein tyrosine phosphatases, contains two tandem SRC homology (SH2) domains at the N-terminus of the catalytic domain. Based on crystallographic studies, the SH2 domain is believed to function as a conformational switch which can bind and inhibit the phosphatase or bind phosphoprotein and activate the enzyme. SHP2 is thought to link growth factor receptors and other signalling proteins via protein-tyrosine phosphorylation. It may play a role in mammalian budding morphogenesis and immune response. Missense mutations in the SHP2 gene have been linked to Noonan Syndrome. Additionally, SHP2 is present in diverse neurons and is thought to play a role in abroad spectrum of neuronal responses, including neuronal survival.
