Antibody Usage Summary: Immunohistochemistry: LS-B2541 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2541 was determined to be 10 ug/ml. Suitable for use in Western Blot and immunoprecipitation.: Western Blot: 0.1-1 ug/ml. Positive control: HeLa cells; Jurkat cells.

Antibody Summary:
Proline-Rich AKT Substrate (PRAS40) is a 40kD substrate of AKT. Activated AKT phosphorylates PRAS40 on threonine 246, enabling PRAS40 to bind to 14-3-3. AKT and PRAS40 are components of the phosphoinositide 3-kinase (PI3K) pathway. This pathway plays a role in glucose uptake, cell growth and apoptosis inhibition. The precise function of PRAS40 is not yet known; however, it has been hypothesized that PRAS40 interacts with SH3 and WW domain containing proteins and may change the function of these proteins. Suitable for use in Western Blot, Immunohistochemistry (FFPE), and immunoprecipitation. Other applications not tested. Recognizes human PRAS40. Reacts with a 40kD band. This band is absent from PRAS40 siRNA-treated HeLa cell extracts. Does not cross-react with mouse.
