Specificity and Use

PTPN11 / SHP-2 / NS1 antibody was raised against synthetic phosphopeptide corresponding to human SHP2 around the phosphorylation site of tyrosine 580 (R-V-YP-E-N).

Recognizes human SHP2, phosphorylated at (Tyr580). Species cross-Reactivity: mouse, rat.
Suitable for use in Immunohistochemistry:1:50-1:100.

SHP2, a NT2 non-receptor protein tyrosine phosphatases, contains two tandem SRC homology (SH2) domains at the N-terminus of the catalytic domain. Based on crystallographic studies, the SH2 domain is believed to function as a conformational switch which can bind and inhibit the phosphatase or bind phosphoprotein and activate the enzyme. SHP2 is thought to link growth factor receptors and other signalling proteins via protein-tyrosine phosphorylation.
