Specificity and Use

DKK1 antibody was raised against synthetic 16 amino acid peptide from N-terminus of human DKK1. Percent identity with other species by BLAST analysis: Human, Gorilla, Monkey, Marmoset, Horse, Pig (100%); Gibbon, Mouse, Rat, Bat, Elephant (94%); Bovine, Hamster (88%); Panda, Rabbit (81%).

Human DKK1. BLAST analysis of the peptide immunogen showed no homology with other human proteins.
Immunohistochemistry: LS-A2865 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-A2865 was determined to be 20 ug/ml.

Dickkopf Homolog 1 (DKK1) is a member of the Dickkopf family. It is a secreted protein with two cysteine rich regions and is involved in embryonic development through its inhibition of the WNT signaling pathway. Elevated levels of DKK1 in bone marrow plasma and peripheral blood is associated with the presence of osteolytic bone lesions in patients with multiple myeloma.
